The phosphate- and Tris-buffered saline formulations are used for blocking excess binding sites on membranes and microplates in antibody-based detection methods. The term casein, as will be used in this report, refers to that mixture of proteins which is obtained by the method of acid precipitation. Casein prepared by other methods will be so designated, where necessary. The method of Hammarsten is the best known of the earlier methods for the laboratory preparation of casein (2).
